Effect of Curcumin on the MMP-1 Expression and Collagen Production in Human Dermal Fibroblast

Abstract
The effect of curcumin, a main component of Curcuma longa , on the MMP-1 and collagen production in human dermal fibroblast was studied (to observe the availabilty for) as a raw material of cosmetics. The effect of curmumin has found that expression of MMP-1 was reduced at lower concentration of curcumin by using HDF cells that UVB (100 mJ/cm©÷) was investigated. Enhanced collagen production was confirmed by repeated tests. Because curcumin affect JNK and p38 and plays the role of hindering PI3K as collagen increases at curcumin concentration of 10 to 11 ¥ìM and revelation of collagen is increased to in the group. Actually, the wstern blot result has found that phosphorylation of JNK was increased when 10 ¥ìM of curcumin concentration was treated. AKT has found that phosphorylation of AKT decreased as the downstream gene of PI3K. It is thought that these results are the same one that collagen production is increased as curcumin affect JNK and curcumin plays the role of PI3K inhibitors depending on concentration. On the other hand, the decrease of collagen production at the higher concentration is not clear yet and further studies are necessary (to elucidate the reason).
